Synthesis Method

Preparation method 1 of 1-Bromo-3,5-difluorobenzene

(1) Add 129kg 2,4-difluoroaniline and 200-500kg dichloroethane to the reactor, stir and cool down, and control the temperature at 10-15℃. Slowly add 85kg of bromine, and after the addition, raise the temperature to 25℃. Add 45-65kg of hydrogen peroxide, and after the addition, react at a constant temperature, and track the reaction process by GC. After 25- 40 minutes, the reaction is complete, and the reaction is stopped, separated, and the organic phase is reserved. The water layer is neutralized and concentrated to obtain sodium bromide.

(2) Add 500kg of hydrochloric acid with a mass concentration of 10% in the reactor in advance, start stirring, and then add 508kg of the organic phase obtained in step (1), and cool down to 0-5℃. Add 67.5kg solid sodium nitrite, and after the addition, use starch potassium iodide test paper to check whether the reaction is complete. After the reaction is completed, add 20-50kg of catalyst and 110kg of sodium hypophosphite. After the addition is completed, keep warm for 30- 50 minutes. Slowly heat to 15-20℃, stir the reaction, and track the reaction progress by GC. After 0.5-3 hours, the reaction is complete, stratified, and distilled to obtain solvent and product.

Preparation method 2 of 1-Bromo-3,5-difluorobenzene

  1. Add a solution of bromobenzene (120 mL, 1.14 mol) in ether (200 mL) to a suspension (100 mL) of effectively stirred magnesium chips (27.8g, 1.16 mol) in dry ether, and maintain gentle reflux under nitrogen after the Grignard formation begins.
  2. After the addition is completed, reflux the mixture for 1-2 hours.
  3. After the mixture is cooled to 0℃, add a solution of dichlorodimethylsilane (130mL, 1.07 mol) in ether (250mL) as soon as possible.
  4. Then reflux the mixture and stir rapidly for 48 hours.
  5. After the mixture was cooled to room temperature, pentane (500 mL) was added, filtered (diatomaceous earth), and the solvent was evaporated in vacuo.
  6. Dimethylbenzene was obtained as a colorless liquid by distillation.
