Application
1-Phenyl-2-pyrrolidone is a pyrrolidone compound. As an important class of nitrogen-containing heterocyclic skeletons, pyrrolidone is widely present in natural products.
Medical field:
1-Phenyl-2-pyrrolidone is a GABA analogue with sedative effects.
Intravenous injection of 50-100 mg/kg can reduce the exploratory behavior of rats and inhibit the emotional response of dogs and cats.
In addition, it can reduce the pressor response to emotional stress, but does not affect the normal reflex function of pressure receptors.
Industrial application:
Ink dispersant: The copolymer formed with butadiene and styrene is used as an ink dispersant to improve the dispersion effect and stability of the ink.

Synthesis Method
1. Method using N-phenylcyclohexylamine, acetonitrile, anhydrous copper acetate, potassium iodide and potassium persulfate complex:
Add N-phenylcyclohexylamine (0.5mmol, 81mg), acetonitrile (5mL), anhydrous copper acetate (1mmol, 181mg), potassium iodide (1mmol, 166mg) and potassium persulfate complex (Oxone, 615mg, 1mmol) to a 10mL Shrek tube in sequence, evacuate and fill with oxygen (1atm), place it in an 80℃ oil bath and stir for 12h.
Then, add 10mL saturated brine to quench the reaction, extract with ethyl acetate (10mL×3), combine the organic phases, and dry with anhydrous sodium sulfate.
Filter, spin dry, and separate through a silica gel column (petroleum ether/ethyl acetate=3/1) to obtain a white solid product 1-phenyl-2-pyrrolidinone (47mg, 58%).
2. Method using 4,4′-diamino-2,2′-bipyridine, trifluoromethanesulfonate manganese (II), hydrogen peroxide and sodium sulfite:
5.59 mg (0.03 mmol) 4,4′-diamino-2,2′-bipyridine (Ligand), 5.30 mg (0.015 mmol) trifluoromethanesulfonate manganese (II) (Mn (OTf)2), and 1.5 mL acetone were added to the reaction tube, stirred at 70 ° C for 10 min, and then cooled to room temperature.
0.5 mmol) 1-phenylpyrrolidine was added, and then 250 μL of 30% hydrogen peroxide was diluted with 0.5 mL acetone and added to the reaction tube continuously for 60 minutes through a flow injection pump.
After the addition, saturated sodium sulfite was added to the reaction solution to quench the remaining hydrogen peroxide, and extracted with ethyl acetate to obtain 1-phenylpyrrolidone-2-one with a yield of 86%.
3. Method using γ-butyrolactone and aniline:
γ-butyrolactone and aniline are transported into the pipeline through a metering pump for mixing, and deionized water is used as a reaction catalyst to react under pressure conditions.
The reaction product is condensed in an ice-water bath to remove heat and then collected to obtain 1-phenyl-2-pyrrolidone.
This method is highly selective and environmentally friendly, and the process operation is simple.
4. Method using phosphoric acid as a catalyst:
γ-butyrolactone and aniline are reacted under the catalysis of phosphoric acid, and the optimal process conditions are n(γ-butyrolactone)∶n(aniline)∶n(phosphoric acid)=1∶1.5∶0.1, the reaction time is 2 hours, and the reaction temperature is 180~210℃.
The product is separated and purified by vacuum distillation and recrystallization, with a yield of 96.2% and a mass fraction of 99.5%.
