1-Fluoronaphthalene Application, Use, Usage, Synthesis Method, etc.

Application:

1. 1-Fluoronaphthalene as a pharmaceutical intermediate, organic synthesis reagent

1-Fluoronaphthalene is a key intermediate of duloxetine.

Duloxetine is an antidepressant with dual inhibitory effects on the reabsorption of serotonin and norepinephrine. It can treat depression, stress incontinence and pain, and is used to treat diabetes.

2. Used in the production of APIs in the pharmaceutical industry

Fluorinated intermediates have the characteristics of high efficacy and low side effects in pharmaceutical synthesis, and can adjust parameters such as electronics and lipophilicity, making the drug performance superior.

3. 1-Fluoronaphthalene is also used in the petroleum industry and other chemical synthesis fields. Its strong stability and physiological activity make it important in many industrial fields.

1-Fluoronaphthalene Synthesis Method

The main fluorination methods on aromatic rings are:

  1. Fluorination is carried out directly with common fluorination reagents or under catalytic conditions.
  2. Some special fluorination reagents, such as patents CN14M195A and US4394527, were used.
  3. Use fluorine to replace other halogens for fluorination.
  4. The method of replacing amino groups by diazotization reaction, such as patent CN1887833.

Detailed introduction of a synthesis method

1 Fluoronaphthalene Synthesis Method Roadmap

1. Diazotization reaction:

Add 550g of hydrochloric acid solution (mass concentration is 20%) and 100g of naphthylamine to a 1000 mL three-necked flask, stir and heat to 75℃ to dissolve, cool to below 5℃, and slowly add 50g of sodium nitrite at this temperature. After adding, stir at a low temperature for 0.2 hours to obtain a diazonium salt solution.

2. Substitution reaction:

Add 150g of fluoroboric acid solution (mass concentration is 40%) to the generated solution obtained in step 1, stir for 0.25h, filter, and dry the filter cake at 65℃ for 0.2h to obtain dry naphthylamine diazonium fluoroboric acid complex salt.

3. Hot air decomposition:

The dried diazonium salt fluoroborate complex salt is slowly added into a reactor with hot air (hot air temperature is 85-90℃), and the dried powdered naphthylamine diazonium salt fluoroborate complex salt is blown up and dispersed by the hot air and absorbs heat for thermal decomposition to obtain a 1-fluoronaphthalene solution containing a small amount of solid impurities.

4. Purification treatment:

The 1-fluoronaphthalene solution obtained in step 3) is first washed with pure water for 3-6 times, then neutralized with soda ash to a pH of 6.8-7.2, and finally filtered to separate the oil layer, and the filtrate is distilled to obtain 69g of naphthalene-based fluorinated intermediate 1-fluoronaphthalene with a content of 99.6%.
