- Starting material for 2-chloropropionamide derivatives used as protein disulfide isomerase (PDI) inhibitors.
- …
Synthesis Method
Method 1: Under the action of acyltransferase, DL-acetylalanine is converted into a-chloropropionic acid by the Fischer method, and L-chloropropionic acid and D-chloropropionic acid are separated by Podbielniak Column distillation.
Method 2: The racemate is split by the lipase of columnar Candida albicans, and the corresponding racemic acid is prepared by enzymatic esterification. One of the enantiomers is esterified and the other remains as an acid. Then, the optically active compounds are separated by distillation or extraction, and D- or L-isomers with a concentration of 70% to 95% can be obtained.
Method 3: Using L-lactic acid as the starting material, R-(+)-2-chloropropionic acid is obtained by esterification, chlorination, and hydrolysis.
